Abstract

This study aims to comprehensively evaluate the chemical composition and biological activities of the essential oil extracted from the seeds of Algerian anise (Pimpinella anisum L.). The essential oil was isolated via hydrodistillation with a yield of 0.96%. Gas Chromatography-Mass Spectrometry (GC-MS) analysis revealed the presence of 31 chemical compounds, with an absolute dominance of trans-anethole (74.4%), followed by estragole (6.3%) and caryophyllene oxide (4.3%). In vitro assays demonstrated outstanding antioxidant activity, with an half-maximal inhibitory concentration (IC50) value of 4.38 mg/mL in the DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging assay, and a high inhibitory capacity against the α-Amylase enzyme, reaching 85.54% at a concentration of 10 mg/mL. Furthermore, the oil exhibited notable anti-inflammatory activity by preventing bovine serum albumin (BSA) denaturation, along with potent antibacterial activity against Escherichia coli and Staphylococcus aureus, while proving its biosafety and non-toxicity toward human erythrocytes at therapeutic concentrations. In the in vivo study, oral administration of the essential oil (100 mg/kg) to diabetic mice led to a significant reduction in fasting blood glucose levels (from 1.12 to 0.45 g/L) and glycated hemoglobin (HbA1c) (from 14.10 to 6.91 mmol/mol), while successfully restoring liver functions by lowering ALT and AST enzyme levels. The in silico molecular docking simulation further supported these findings, revealing that caryophyllene oxide exhibited a binding energy (–5.88 kcal/mol) that outperforms the standard drug metformin in inhibiting the α-Amylase enzyme, alongside confirming ideal pharmacokinetic profiles (ADMET) for the major compounds. In conclusion, this study demonstrates that Algerian anise represents a promising natural source for developing multi-target therapeutic agents for diabetesتهدف هذه الدراسة إلى التقييم الشامل للتركيب الكيميائي والفعالية البيولوجية للزيت الأساسي المستخلص من بذور نبات اليانسون الجزائري (Pimpinella anisum L).
