Abstract
HSA.
This study focuses on evaluating the antioxidant activity of two natural aliphatic reference
compounds—gallic acid (3,4,5-trihydroxybenzoic acid, PubChem CID 370) and ascorbic acid
(vitamin C, PubChem CID 155903693)—using the DPPH radical method, as well as on the
quantitative analysis of their pharmacological synergy in combination. A comprehensive dose-
response matrix (concentrations: 0, 12.5, 25, 50, and 100 μM) was constructed for both
compounds as monotherapies and in combination, then analyzed using the SynergyFinder 3.0
platform, applying four mathematical models: Bliss Independence, Loewe Additivity, ZIP, and
HSA. The overall synergy scores obtained were −13.12 (Bliss), −2.26 (HSA), +1.76 (Loewe),
and −12.35 (ZIP), respectively, revealing an interaction that was generally additive to slightly
antagonistic. In parallel, molecular docking was performed on the target protein 1N8Q using
the CB-Dock2 server. The best affinity scores were −6.6 kcal/mol for gallic acid and −6.1
kcal/mol for ascorbic acid. This thesis contributes to the understanding of molecular interaction
mechanisms at the interface of organic polyphenol chemistry and computational pharmacology.
